My drug screen was part of BOTH DOT physicals I did. So a better answer is: depends. On state and who is conducting the exam.
However, it is not required by either FMCSA Regs or State Laws to obtain a CDL.
As mentioned previously, if the physical was part of a Company Physical - it would (likely) include a drug screen. If you just walk into a clinic to get a DOT Phys, a drug screen would not be part of it, unless requested by a company.

The only times I have had drug screens done at the same time as a physical was at orientation or starting school. If you didn't fill out chain of custody paperwork then it was just a blood/sugar/protein check.
